Az on-line vásárlás vevői elfogadottsága Magyarországon (Adoption of on-line commerce in Hungary)
A hazai internethasználat terjedésével egyre népszerűbbek az on-line kereskedelmi modellek, egyre több
cég kísérli meg, hogy terméket vagy szolgáltatást értékesítsen az interneten. Például az on-line könyvértékesítések
és a diszkont légitársaságok jegyeladásai az e-kereskedelem kiugró ágazatai közé tartoznak
– ez a hasonlóság adja a tanulmány alapját. Dinamikusan növekvő keresleti igények honorálják
az internetes megjelenést, és fordítva: a fogyasztói e-felkészültséget gyakran a kínálati oldal indukálja.
A szerzők kutatásukban a hazai internethasználat bemutatásával és fókuszcsoportos beszélgetések révén
próbálják meg feltárni, hogy mi minden szükséges ahhoz, hogy valódi alternatívát jelentsen az on-line
kereskedelem a vevők számára. Kísérletet tesznek a keresleti oldal átfogó bemutatására, hasonlóságokra
és különbségekre fókuszálva, valamint szintén összevetik a kínálati oldal jellemző karaktereit, ezek egyezőségeit
és eltéréseit vizsgálva. ______________ With the spreading of Hungarian internet usage online
e-commerce models have become more and
more popular, more and more companies start to sell
products and services via the internet. For instance the
on-line book sales and purchase of discount airline
tickets belong to the leading branches of e-business
and demonstrate that dynamically increasing demand
generates supply. In this research first the authors
introduces the basic data about internet usage in Hungary.
Then, based on a series of focused group discussions
they examine what the key customer requirements are
to really consider e-commerce as a serious alternative
for traditional shopping. They attempt to describe the
demand side and juxtapose it with the characteristics of
the supply side of Hungarian e-commerce
Prealbumin epitópok haptén hordozó szerepének vizsgálata limfocita transzformációs tesztben a gyógyszerallergia igazolására = The role of prealbumin as a hapten-carrier protein in lymphocyte transformation test for the establishment of drug allergy
Korábbi vizsgálataink alapján célul tűztük ki, hogy megvizsgáljuk a prealbumin egyes szerkezeti elemeinek haptén hordozó szerepét és különböző gyógyszerekhez kapcsolva nagyszámú betegben meghatározzuk, növelhető-e a limfocita transzformációs teszt (LTT) diagnosztikus hatékonysága. 119 biztosan gyógyszer érzékenységben szenvedő beteg csak gyógyszerrel ill. gyógyszer-prealbumin peptid konjugátummal végzett párhuzamos LTT vizsgálata során nem találtunk konzekvens különbséget a gyógyszer-peptid javára az LTT vizsgálat érzékenységében. Gyógyszeraktiválást követően a különböző immunglobulinok (IgD, IgM, IgG) sejtfelszíni kifejeződése individuálisan változott a betegek B limfocitáin. 84 gén expresszióját vizsgáltuk 2 egészséges egyén és 4 gyógyszerallergiában szenvedő beteg izolált limfocitáin a gyógyszerindukciót követően, amely gének a CD4+ helper T limfociták TH1-TH2 típusaival voltak kapcsolatban. A betegek esetében az érzékenyítő gyógyszer hatására sokkal több vizsgált gén kifejeződésének növekedése következett be. Az is megállapítható volt, hogy a TH1 és TH2 típusú válaszok egyaránt indukálódnak a gyógyszerallergiás betegeknél, bár egyénenként különböző kifejeződési mintázatot mutattak. Eredményeink megerősítik azt a vélekedést, hogy a gyógyszerallergia mind klinikai megjelenésében és lefolyásában, mind immunológiai mechanizmusaiban heterogén kórkép, a TH1 és TH2 típusú válaszok keverednek. | Human prealbumin acting as a hapten-carrier protein in drug allergy was suggested by our previous investigations. The aim of the work was that different structural elements of prealbumin acting as hapten-carrier peptides, in conjugation of different drugs, are able to increase the diagnostic sensitivity of lymphocyte transformation test (LTT) in drug allergy patients comparing to simple drugs. Gene expressions of drug-specific lymphocytes during drug activation were also investigated. Unfortunately, drug-prealbumin conjugates as activators were not able to increase the diagnostic sensitivity of LTT test comparing to simple drug activation based on detailed parallel measurements in 119 patients with proved drug allergy. Immunoglobulin expressions (IgD, IgM, IgG) on the drug-induced B lymphocytes of the patients are individually varied compared to freshly isolated B lymphocytes of the patients as well as to healthy controls.We investigated the expressions of 84 genes (in connection with TH1 and/or TH2 immune response) in freshly separated and drug-induced lymphocytes isolated from healthy controls and patients with drug allergy. The expressions of a lot of genes are increased in the drug-induced lymphocytes of drug allergy patients, however, their expression patterns are varied individually. Our results supported that drug allergy is a heterogeneous disorder in its clinical picture and immunological mechanisms. Both TH1 and TH2 immune responses are involved in the pathogenesis

Visualization of Calcium Dynamics in Kidney Proximal Tubules
Intrarenal changes in cytoplasmic calcium levels have a key role in determining pathologic and pharmacologic responses in major kidney diseases. However, cell-specific delivery of calcium-sensitive probes in vivo remains problematic. We generated a transgenic rat stably expressing the green fluorescent protein-calmodulin-based genetically encoded calcium indicator (GCaMP2) predominantly in the kidney proximal tubules. The transposon-based method used allowed the generation of homozygous transgenic rats containing one copy of the transgene per allele with a defined insertion pattern, without genetic or phenotypic alterations. We applied in vitro confocal and in vivo two-photon microscopy to examine basal calcium levels and ligand- and drug-induced alterations in these levels in proximal tubular epithelial cells. Notably, renal ischemia induced a transient increase in cellular calcium, and reperfusion resulted in a secondary calcium load, which was significantly decreased by systemic administration of specific blockers of the angiotensin receptor and the Na-Ca exchanger. The parallel examination of in vivo cellular calcium dynamics and renal circulation by fluorescent probes opens new possibilities for physiologic and pharmacologic investigations
Differential expression of D-type cyclins in HaCaT keratinocytes and in psoriasis
In this study, we show that the G(0)-G(1)/S phase of HaCaT keratinocyte
cell cycle is characterized by D1-type cyclin expression, whereas
during the repeated rapid turnover of highly proliferating cells, the
expression of cyclins D2 and D3 dominates. Knocking down cyclin D1 mRNA
resulted in no change of cell proliferation and morphology, indicating
that D2 and D3 cyclins could substitute for D1 in driving the cell
cycle. Increased numbers of cyclin D1-expressing keratinocytes were
found in the basal layers of the lesional psoriatic epidermis compared
to both normal and non-lesional epidermis without increased expression
of cyclin D1 mRNA, suggesting a possible dysfunction in the degradation
of cyclin D1 protein. We also detected a significant increase in cyclin
D2 and D3 mRNA expressions in psoriatic epidermis compared to normal
epidermis with no difference in protein expressions. Blocking alpha
5-integrin function by a neutralizing antibody in HaCaT keratinocytes
downregulated the expression of cyclin D1 mRNA without affecting the
expressions of cyclin D2 and D3 indicating a regulatory role for alpha
5-integrin in the expression of cyclin D1. Our data suggest a possible
role for D-type cyclins in the excessive basal-cell proliferation and
perturbed keratinocyte differentiation in the psoriatic epidermis
